Mr. Sack joined D.E. Shaw on Monday in the new position, confirmed spokeswoman Lonna Leong in an interview.
Mr. Sack was executive vice president and a special adviser to the president of the Federal Reserve Bank of New York.
Mr. Sack's co-director is Angel Ubide, also a senior vice president, who joined D.E. Shaw in January from Tudor Investment, where he served as director of global economics.
Messrs. Sack and Ubide report to Max Stone, managing director and head of the company's macro trading strategies group.
D.E. Shaw managed $28 billion in a range of quantitative hedge fund, credit and long-only investment strategies as of Jan. 1.
